Phosphorylated TDP-43 (pTDP-43) aggregates are a signature of amyotrophic lateral sclerosis (ALS). Microscopy-guided automated photo-biotinylation at the locations of pTDP-43 of a postmortem FFPE brain section of an ALS patient enables the subsequent spatial purification and proteomic discovery, providing a novel list of protein constituents at the TDP-43 aggregates of ALS.
